Overview

MW-401 is an orally bioavailable small molecule antagonist and inverse agonist of G-protein-coupled receptor 75 (GPR75), currently in the discovery stage for the treatment of obesity and chronic kidney disease (CKD). GPR75 is an orphan receptor expressed in hypothalamic neurons, adipose tissue, liver, kidney, and vascular endothelium; genetic studies have linked loss-of-function variants of this receptor to lower body mass index (BMI) and protection from obesity and type 2 diabetes. Developed by Mwyngil Therapeutics, MW-401 was in-licensed from Expert Systems Inc. in November 2025. Preclinical studies have demonstrated that oral dosing of MW-401 significantly reduces weight gain in diet-induced obesity models while sparing lean body mass. Additionally, GPR75 modulation has been shown to alleviate filtration stress in the kidney, suggesting potential for kidney protection. First-in-human clinical trials for the program are anticipated to begin in 2027.
