MW-504 is a highly potent, selective, and orally bioavailable small-molecule inhibitor of the NLRP3 (NACHT, LRR and PYD domains-containing protein 3) inflammasome. Developed by Mwyngil Therapeutics, the drug is currently in the IND-enabling stage for the treatment of obesity and chronic kidney disease (CKD). By inhibiting the NLRP3 inflammasome, MW-504 prevents the activation of caspase-1 and the subsequent maturation and release of pro-inflammatory cytokines such as IL-1β and IL-18. These cytokines are key mediators of the chronic low-grade inflammation that drives the progression of metabolic disorders and renal fibrosis, making NLRP3 an attractive target for therapeutic intervention in obesity-related complications and CKD.
