Overview

MW-601 is an orally bioavailable, brain-penetrant small molecule inhibitor of protein-tyrosine phosphatase 1B (PTP1B) under development by Mwyngil Therapeutics for the treatment of obesity and cardiometabolic disorders. PTP1B acts as a negative-feedback regulator of leptin and insulin signaling in the hypothalamus. In obese states, PTP1B is overexpressed, leading to hypothalamic resistance to these satiety and energy-sensing signals. By crossing the blood-brain barrier and inhibiting PTP1B, MW-601 aims to restore leptin and insulin sensitivity (hypothalamic re-sensitization), thereby promoting POMC neuron activation to suppress appetite and inhibiting AgRP/NPY neurons to reduce hunger drive. The candidate is currently in Phase 1 clinical development.
