MW-C1 is a novel antibody-drug conjugate (ADC) designed to target Claudin 1 (CLDN1), a tight junction protein frequently overexpressed in solid tumors such as head and neck squamous cell carcinoma (HNSCC), lung squamous cell carcinoma (LUSC), and hepatocellular carcinoma (HCC). Developed by Mabwell Bioscience, MW-C1 utilizes site-specific interchain-disulfide conjugation technology to attach the cytotoxic payload monomethyl auristatin E (MMAE) to a CLDN1-specific antibody via a cleavable linker, maintaining a drug-to-antibody ratio (DAR) of 4. Preclinical evaluations in cell-line-derived and patient-derived xenograft models have demonstrated significant anti-tumor efficacy and durable tumor regression. Additionally, non-human primate studies indicate a favorable safety profile and pharmacokinetic properties, supporting its potential as a therapeutic candidate for CLDN1-positive malignancies.
