MW11-h317 is a humanized monoclonal antibody that targets the Programmed cell death 1 (PD-1) receptor, an immune checkpoint protein. It is distinguished from first-generation PD-1 inhibitors, such as nivolumab and pembrolizumab, by its unique binding epitope that is dependent on the N-linked glycosylation of PD-1, specifically at the Asn58 residue. By binding to this glycosylated site, MW11-h317 effectively blocks the interaction between PD-1 and its ligands, PD-L1 and PD-L2, thereby reversing T-cell immunosuppression and promoting an antitumor immune response. Preclinical evaluations in mouse models have shown that MW11-h317 induces T-cell-mediated immune responses and inhibits tumor growth with potency comparable to established clinical antibodies.
