Overview

MW151 is an orally bioavailable, brain/CNS-penetrant small molecule with anti-inflammatory and neuroprotective properties. It was discovered by screening compounds that inhibit proinflammatory cytokine production in activated glial cells. The drug selectively suppresses the overproduction of proinflammatory cytokines—such as interleukin 1 beta (IL‑1β)—without affecting basal levels or blocking anti-inflammatory cytokines. This homeostatic modulation of neuroinflammation is thought to protect against synaptic dysfunction and neuronal loss associated with diseases like Alzheimer’s disease (AD), traumatic brain injury (TBI), multiple sclerosis (MS), seizure disorders, and radiation-induced cognitive impairment. The precise molecular target remains undisclosed but may involve inhibition of induced glycolysis in myeloid cells during inflammatory activation[1][2][3][4][5][6][8]. Developed by ImmunoChem Therapeutics and originally at Northwestern University labs, MW151 is currently in Phase 1 clinical trials for Alzheimer’s disease.
