MX006 is an antibody-drug conjugate (ADC) targeting B7-H3 (CD276), developed by Myricx Pharma. It is the first clinical candidate to utilize Myricx's proprietary N-myristoyltransferase inhibitor (NMTi) payload platform. N-myristoyltransferase is an enzyme responsible for the co-translational and post-translational covalent attachment of a myristoyl group to the N-terminal glycine of various proteins, a process essential for the membrane localization and functional activity of many proteins involved in oncogenic signaling and survival. By delivering a potent NMT inhibitor specifically to B7-H3-expressing cancer cells, MX006 aims to induce cell death by disrupting multiple essential cellular pathways simultaneously. B7-H3 is a transmembrane protein and member of the B7 family of immune checkpoint molecules that is frequently overexpressed in a wide range of solid tumors, including lung, prostate, and breast cancers, while maintaining low expression in normal tissues. MX006 is currently being investigated in a Phase 1 first-in-human clinical trial for patients with advanced or metastatic solid tumors known to express B7-H3.
