MX35 is a murine monoclonal antibody that specifically targets the sodium-dependent phosphate transport protein 2b (NaPi2b), also known as SLC34A2. This antigen is highly expressed on the surface of epithelial ovarian cancer cells and in normal lung tissue (type II pneumocytes), but has limited expression elsewhere, making it a viable target for directed therapy. Originally developed at Memorial Sloan Kettering Cancer Center, MX35 has been extensively studied as a targeting vehicle for alpha-radioimmunotherapy. In this context, it is conjugated to alpha-emitting radioisotopes such as Bismuth-213 (^213Bi) or Astatine-211 (^211At) to deliver high-linear energy transfer (LET) radiation directly to micrometastatic disease, particularly within the peritoneal cavity in patients with ovarian cancer. While the murine version has been used in early-phase clinical trials, humanized versions (hMX35) have also been developed to reduce immunogenicity.
