My96CAR is a chimeric antigen receptor T cell (CAR-T) construct engineered to target the CD33 antigen, primarily expressed on acute myeloid leukemia (AML) cells. The construct is based on a single-chain variable fragment (scFv) derived from the My96 antibody, which specifically binds to an epitope in the V domain of the full-length CD33 (CD33M) isoform. In preclinical models, My96CAR T cells demonstrated significant and specific cytotoxicity against CD33^high^ leukemia cell lines (such as HL-60 and MOLM-14), with little to no toxicity toward CD33^low^ or CD33-negative lines, indicating selective anti-leukemic activity[1][5]. My96CAR induced pro-inflammatory cytokine production and was efficacious in eliminating CD33-positive leukemic cells both in vitro and in vivo[5]. The design and evaluation of My96CAR provided a reference or positive control in studies developing next-generation CAR T cells with fully human immunoglobulin heavy-chain targeting domains[1][5].
