MYC c-mod is an oral small molecule condensate modulator (c-mod) developed by Dewpoint Therapeutics to target the MYC oncoprotein. MYC is a transcription factor that is frequently amplified or overexpressed in various human cancers but has traditionally been considered undruggable due to its intrinsically disordered structure and lack of stable binding pockets. This therapeutic approach utilizes Dewpoint's condensate science platform to identify molecules that selectively dissolve MYC nuclear condensates, thereby disrupting its oncogenic transcriptional activity and repressing the expression of downstream target genes. Preclinical data presented at the 2025 AACR-NCI-EORTC conference demonstrated that these c-mods induce cell cycle arrest and apoptosis in various solid tumor models. In vivo studies in ovarian and esophageal cancer xenografts showed that oral administration of MYC c-mod leads to tumor stasis or regression, supported by a clear pharmacokinetic/pharmacodynamic relationship in reducing MYC protein levels.
