MYC siRNA refers to small interfering RNA (siRNA) molecules designed to target and silence the expression of the MYC oncogene (specifically c-MYC mRNA) via the RNA interference (RNAi) pathway. MYC is a highly sought-after but historically 'undruggable' transcription factor that is frequently overexpressed or deregulated in a wide variety of human cancers, driving cell proliferation, metabolic reprogramming, and therapeutic resistance. By binding complementarily to MYC mRNA, MYC siRNA guides the RNA-induced silencing complex (RISC) to cleave and degrade the transcript, thereby reducing MYC protein levels. Various delivery vehicles, such as lipid nanoparticles (LNPs), are utilized to protect the siRNA from nuclease degradation and facilitate intracellular delivery. While several preclinical and clinical-stage candidates (such as Dicerna's DCR-MYC) have been developed, targeting MYC via siRNA remains an active area of oncology research.
