Overview

This is a combination therapy of mycophenolate mofetil and obinutuzumab, currently in late-stage clinical development for the treatment of class III or IV lupus nephritis (LN), a severe manifestation of systemic lupus erythematosus. Mycophenolate mofetil is an immunosuppressant that acts as a reversible inhibitor of inosine monophosphate dehydrogenase, suppressing T and B lymphocyte proliferation. Obinutuzumab is a humanized type II anti-CD20 monoclonal antibody that binds to the CD20 antigen on B cells, leading to their depletion. The combination aims to enhance immunosuppression by targeting both lymphocyte proliferation (mycophenolate) and B cell-mediated autoimmunity (obinutuzumab). This regimen has shown improved renal responses compared with standard-of-care therapies in phase 2 trials and is being further evaluated in global phase 3 studies for lupus nephritis[1][4][8][9].
