Overview

This entry refers to a combination of three immunosuppressive drugs: mycophenolate mofetil, mycophenolate sodium, and azathioprine. All are used primarily to prevent organ transplant rejection and treat various autoimmune diseases. Mycophenolate mofetil and mycophenolate sodium are prodrugs of mycophenolic acid, which inhibits inosine monophosphate dehydrogenase (IMPDH), leading to suppression of lymphocyte proliferation by blocking de novo guanosine nucleotide synthesis. Azathioprine is a purine analog that interferes with DNA synthesis in rapidly dividing cells, particularly affecting T and B lymphocytes. Both drug types reduce immune system activity but via different mechanisms. They are commonly used as steroid-sparing agents in conditions such as lupus nephritis, rheumatoid arthritis (azathioprine), kidney transplantation (all three), bullous pemphigoid, and other autoimmune disorders[1][2][5].
