Graftide-M/S is a brand name used by Viatris (formerly Mylan) in India to represent its line of mycophenolate-based immunosuppressive medications, which include Graftide-M (containing mycophenolate mofetil) and Graftide-S (containing mycophenolate sodium). Both formulations act as prodrugs that are rapidly metabolized in the body to the active moiety, mycophenolic acid. Mycophenolic acid is a potent, selective, and reversible inhibitor of inosine-5'-monophosphate dehydrogenase (IMPDH), a key rate-limiting enzyme in the de novo synthesis of guanosine nucleotides. Because T and B lymphocytes are highly dependent on de novo purine synthesis for proliferation, whereas other cell types can utilize salvage pathways, mycophenolic acid selectively suppresses lymphocyte proliferation and antibody formation. Graftide-M/S is primarily indicated for the prophylaxis of organ rejection in patients receiving allogeneic renal, cardiac, or hepatic transplants, typically administered in combination with other immunosuppressive agents such as cyclosporine and corticosteroids.
