MyD88 siRNA is a small interfering RNA designed to silence the expression of the Myeloid Differentiation Primary Response 88 (MyD88) gene. MyD88 is a critical adapter protein in the Toll-like receptor (TLR) and Interleukin-1 receptor (IL-1R) signaling pathways, which are essential for innate immunity and inflammatory responses. In the context of oncology, particularly melanoma, aberrant MyD88 signaling has been implicated in promoting chronic inflammation, oxidative stress, and tumor cell survival. Preclinical research has demonstrated that siRNA-mediated knockdown of MyD88 in melanoma cell lines leads to significant growth inhibition, induction of autophagy, and a reduction in inflammatory markers such as Cox-2 and reactive oxygen species. This approach is being explored as a potential therapeutic strategy for melanoma patients with high constitutive expression of the IL-1/MyD88 signaling axis.
