Overview

Myeloma Immunoglobulin Idiotype Vaccine is a personalized cancer vaccine that targets the unique idiotype (Id) determinants on the monoclonal immunoglobulin (paraprotein) produced by malignant plasma cells in multiple myeloma. The idiotype, formed through gene rearrangement during B-cell maturation and somatic hypermutation, represents a tumor-specific antigen. The vaccine is designed to induce both humoral and cellular immune responses against these tumor-specific idiotypes, potentially eliminating residual disease after conventional treatments like autologous stem cell transplantation. Various delivery methods have been explored, including DNA-based vaccines, protein conjugates with keyhole limpet hemocyanin (KLH), and dendritic cell-based approaches.
