Drug intelligence / Profile preview
Myr-T3 is a peptide-drug conjugate consisting of Myrcludex B (bulevirtide), a synthetic peptide targeting the liver-specific bile salt transporter sodium taurocholate co-transporting polypeptide (NTCP), chemically conjugated to triiodothyronine (T3), a thyroid hormone [3181]. Developed by Amsterdam UMC and Novo Nordisk, Myr-T3 is designed for liver-specific delivery of T3 to enhance hepatic lipid metabolism and treat metabolic dysfunction-associated steatotic liver disease (MASLD) and hypercholesterolemia, while minimizing the systemic cardiovascular and skeletal toxicities typically associated with thyroid hormone administration [3181]. Preclinical studies have demonstrated that Myr-T3 preserves the biological activity of both components, showing hepatic efficacy in reducing plasma cholesterol and upregulating T3-responsive genes in the liver, though further structural optimization is required to improve its organ specificity and prevent systemic toxicity at high doses [3181].
