Drug intelligence / Profile preview
Myristoylated human defensin 5 (HD5-myr) is a C-terminally myristoylated, self-assembling antimicrobial peptide (AMP) derived from human α-defensin 5 (HD5). Developed by researchers at Zhejiang University and collaborating institutions, HD5-myr self-assembles into nanostructures (nanobiotics) that exhibit enhanced broad-spectrum bactericidal activity compared to unmodified HD5. It functions by disrupting bacterial cell walls and membranes, and by directly binding and neutralizing bacterial endotoxins (lipopolysaccharides, LPS). The resulting HD5-myr/LPS complexes are rapidly cleared by macrophages via F-actin-dependent phagocytosis, which upregulates acyloxyacyl hydrolase (AOAH) expression to facilitate intracellular LPS degradation []. HD5-myr is under preclinical investigation for the treatment of sepsis, bacterial infections (including MRSA skin infections), and as a component of antimicrobial wound dressings.
