Overview
Myristyl nicotinate is a lipophilic small molecule ester prodrug of nicotinic acid (niacin) developed by Niadyne Pharma for topical application. It is designed to penetrate the skin barrier and undergo hydrolysis to release nicotinic acid, which subsequently acts as an agonist for the G protein-coupled receptors HCAR2 (NIACR1) and HCAR3 (HM74). This activation stimulates epidermal differentiation, increases intracellular NAD levels, and enhances skin barrier function. The compound has been investigated in clinical trials for the prevention of actinic keratosis and its progression to skin cancer, as well as for the treatment of chronic photodamage and atopic dermatitis.
