Overview

Myrobalan Therapeutics is developing an oral, brain-penetrant, allosteric small molecule inhibitor of Tyrosine Kinase 2 (TYK2) for the treatment of neuroinflammatory and neurodegenerative diseases. TYK2 is a member of the Janus kinase (JAK) family that mediates signaling for several pro-inflammatory cytokines, including IL-12, IL-23, and Type I interferons. By targeting the allosteric pseudokinase domain rather than the conserved orthosteric catalytic domain, this compound aims for high selectivity over other JAK family members (JAK1, JAK2, JAK3), potentially reducing off-target toxicities. The program is specifically designed to cross the blood-brain barrier to address central nervous system (CNS) pathologies such as multiple sclerosis, Alzheimer's disease, and amyotrophic lateral sclerosis (ALS). As of early 2024, the program is in the discovery and preclinical stages, working toward IND-enabling studies.
