MYTX-011 is a novel antibody-drug conjugate (ADC) designed to target c-MET, a receptor tyrosine kinase implicated in various cancers. It consists of a humanized monoclonal antibody engineered for pH-dependent binding to c-MET, which enhances internalization and delivery of its cytotoxic payload, monomethyl auristatin E (MMAE), into tumor cells. This pH-dependent mechanism allows MYTX-011 to be effective across tumors with varying levels of c-MET expression, potentially expanding its therapeutic reach beyond patients with high c-MET expression. Preclinical studies have demonstrated superior internalization and cytotoxicity compared to non–pH-engineered ADCs and improved pharmacokinetics. The drug is being developed primarily for non-small cell lung cancer (NSCLC) but has shown activity in other solid tumor models expressing c-MET. A first-in-human clinical trial is ongoing in patients with previously treated locally advanced or metastatic NSCLC.
