**MyVax** was a Genitope-developed, patient-specific therapeutic cancer vaccine for B-cell malignancies, principally follicular lymphoma. Each individualized product contained a recombinant form of the patient’s tumor immunoglobulin idiotype, representing the clonal B-cell tumor antigen, chemically conjugated to keyhole limpet hemocyanin to increase immunogenicity. It was administered by subcutaneous injection with locally administered granulocyte-macrophage colony-stimulating factor as an immunostimulatory adjunct. The intended mechanism was induction of tumor idiotype-specific humoral and cellular immune responses against malignant B cells. A randomized phase III trial in previously untreated advanced follicular lymphoma did not improve progression-free survival versus a KLH plus granulocyte-macrophage colony-stimulating factor control, and Genitope subsequently suspended further MyVax development.
