Overview

WGI-0401 (Myzomib) is a liposomal formulation of the proteasome inhibitor bortezomib, currently in preclinical development by The WhiteOak Group for the treatment of multiple myeloma. By encapsulating bortezomib within a lipid nanoparticle, the drug aims to improve the pharmacokinetic profile and therapeutic index of the standard-of-care proteasome inhibitor, potentially reducing systemic toxicities such as peripheral neuropathy while enhancing delivery to tumor cells. It acts by binding to and inhibiting the 26S proteasome, leading to the accumulation of ubiquitinated proteins and subsequent apoptosis in malignant plasma cells. The program is currently in the IND-enabling stage of development.
