MZB1 CAR T cell is an investigational, autologous chimeric antigen receptor T-cell therapy engineered to recognize the intracellular endoplasmic reticulum protein marginal zone B and B1 cell-specific protein (MZB1) when it is presented as a peptide in complex with HLA-A*02:01 on the surface of malignant plasma cells. Using a TCR-like CAR construct, these modified T cells bind MZB1/HLA-A2 complexes and mediate targeted cytotoxicity against MZB1-positive, HLA-A2–restricted multiple myeloma and Waldenström macroglobulinemia cells, with preclinical data demonstrating potent, specific antitumor activity and robust activation marker upregulation and effector cytokine and granzyme release.[2][5][7][9][11]
