Overview

MZe686 is a potent, selective, and orally bioavailable small molecule inhibitor of the Mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1) protease. MALT1 is a paracaspase that plays a critical role in the CBM (CARMA1-BCL10-MALT1) complex, which is essential for B-cell receptor (BCR)-mediated NF-κB signaling. In many B-cell malignancies, particularly activated B-cell-like diffuse large B-cell lymphoma (ABC-DLBCL) and mantle cell lymphoma (MCL), MALT1 protease activity is constitutively active, driving tumor cell survival and proliferation. By inhibiting MALT1, MZe686 disrupts the NF-κB pathway and induces apoptosis in malignant B cells. Developed by MZE Biotech, MZe686 is currently undergoing Phase 1 clinical evaluation for the treatment of relapsed or refractory B-cell non-Hodgkin lymphomas.
