MZE829 is an oral, small molecule inhibitor of apolipoprotein L1 (APOL1), currently in clinical development for the treatment of APOL1-mediated kidney disease (AMKD), also referred to as APOL1 kidney disease (AKD). Genetic variants in the APOL1 gene are associated with increased risk and progression of chronic kidney disease, particularly among individuals of West African ancestry. MZE829 selectively inhibits APOL1, aiming to reduce proteinuria and slow or prevent progression of AKD. The drug has demonstrated favorable safety and tolerability in Phase 1 trials with dose-proportional pharmacokinetics supporting once-daily dosing. It is being developed by Maze Therapeutics, which has initiated a Phase 2 clinical trial to further evaluate its efficacy and safety in patients with AKD[1][2][3][4][5][6][7].
