N(1)-acetyl-N(2)-formyl-5-methoxykynuramine (AFMK) is a primary kynuramine metabolite of melatonin, produced through the oxidative pyrrole-ring cleavage of melatonin by enzymes such as indoleamine 2,3-dioxygenase (IDO) or non-enzymatic reactions with reactive oxygen species. AFMK is a potent antioxidant and broad-spectrum free radical scavenger that protects cells from oxidative stress and mitochondrial dysfunction. Research indicates it possesses anti-inflammatory, neuroprotective, and anti-proliferative properties, showing potential in treating Alzheimer's disease, acute pancreatitis, and skin disorders. It acts partly through signaling via melatonin receptors (MT1 and MT2) and serotonin receptors (5-HT3). AFMK also serves as a precursor to another bioactive metabolite, N(1)-acetyl-5-methoxykynuramine (AMK).
