N-(2-(methylamino)ethyl)-5-isoquinolinesulfonamide (commonly known as H-8) is a small molecule inhibitor of protein kinases. It is primarily used in research settings to inhibit cyclic nucleotide-dependent protein kinases such as cAMP-dependent protein kinase (PKA), with additional activity against other kinases including cGMP-dependent protein kinase (PKG). The compound acts by competitively inhibiting the ATP binding site on the catalytic subunit of these kinases. While it was initially believed to be relatively selective for PKA, subsequent studies have shown that it can also inhibit other serine/threonine kinases at higher concentrations. It has been widely used as a tool compound in cell signaling research but is not approved for clinical use[1][3][7].
