SCH 365351 is a potent and highly selective phosphodiesterase 4 (PDE4) inhibitor, with an IC50 of 20 nM for PDE4[1][5]. It is a major active metabolite of SCH 351591 in rats, mice, and monkeys[5]. Both SCH 351591 and SCH 365351 inhibit cytokine production in human blood mononuclear cells and have shown preclinical activity in animal models of lung inflammation, including attenuation of allergen-induced eosinophilia and airway hyperreactivity in allergic guinea pigs[1]. SCH 365351 is also orally bioavailable and has demonstrated efficacy in animal models of asthma and chronic obstructive pulmonary disease (COPD)[1]. No evidence of human clinical development or approval was found in the available information.
