Overview

**N-803 + atezolizumab + PD-L1 t-haNK** is an investigational combination immunotherapy regimen being studied primarily for advanced and metastatic cancers, including pancreatic cancer and head and neck squamous cell carcinoma. The combination includes: * **N-803**: a recombinant human interleukin 15 (IL‑15) superagonist complex that stimulates the proliferation and activation of natural killer (NK) cells and CD8+ T cells, enhancing anti-tumor immune responses. * **Atezolizumab**: a monoclonal antibody targeting programmed death-ligand 1 (**PD-L1**) to block its interaction with the PD‑1 receptor on T cells, thereby preventing immune evasion by tumor cells. * **PD-L1 t-haNK**: allogeneic, off-the-shelf NK cells genetically engineered to express a chimeric antigen receptor targeting PD‑L1 on tumor cells, enabling direct cytotoxicity against tumors expressing this checkpoint molecule. This multi-modal approach aims to orchestrate both innate and adaptive immunity by combining checkpoint inhibition with cellular immunotherapy. Early clinical reports have shown promising activity in heavily pretreated patients with metastatic cancers[3][4][6][8].
