This is a combination immunotherapy regimen consisting of three agents: **N-803** is a recombinant human interleukin 15 (IL‑15) superagonist complex designed to stimulate the proliferation and activation of natural killer (NK) cells and CD8+ T cells, thereby enhancing antitumor immune responses. **Durvalumab** is a monoclonal antibody that targets programmed death-ligand 1 (PD‑L1), blocking its interaction with the PD‑1 receptor on T cells, which releases inhibition of immune responses against tumor cells. **PD-L1 t-haNK** refers to engineered high-affinity natural killer cells expressing a chimeric antigen receptor specific for PD‑L1, as well as high-affinity CD16 and ER-retained IL‑2. These modified NK cells directly target and kill tumor cells expressing PD‑L1, including those resistant to T cell-mediated killing due to loss of MHC-I expression. The combination aims to maximize both innate and adaptive immune system activity against tumors by stimulating endogenous immunity with N‑803, blocking inhibitory signals with durvalumab, and providing exogenous cytotoxic NK cell activity via PD-L1 t-haNK therapy. This approach has been investigated in advanced solid tumors such as head and neck squamous cell carcinoma and pancreatic cancer[4][5].
