Overview

N-acetyl glucosamine (GlcNAc) is an amino sugar and a precursor for the synthesis of glycosaminoglycans and glycoproteins, which are essential components of the intestinal mucosal barrier. It is being evaluated by Queen Mary University of London in the TAME (Therapeutic Approaches for Malnutrition Enteropathy) trial for the treatment of severe acute malnutrition (SAM) and malnutrition enteropathy in children. The therapeutic rationale is based on the observation that glycosylation is deficient in SAM; providing GlcNAc is intended to restore the intestinal barrier and promote mucosal healing. In a Phase 2 randomized trial conducted in Zambia and Zimbabwe, the intervention was found to be safe and associated with reduced systemic inflammation and diarrhea, although it did not show a significant difference from standard care on the primary endpoint.
