N-arachidonoyl dopamine (NADA) is an endogenous cannabinoid and vanilloid receptor agonist that is being investigated for its potential anti-tumor activity in hematological malignancies. Research presented at ASH 2024 indicates that NADA induces pyroptosis in leukemia cells by directly binding to Gasdermin D (GSDMD) and promoting its cleavage by Caspase-1 (CASP1). Additionally, NADA has been shown to inhibit NRAS membrane translocation and induce oncosis-like cell death. In preclinical models, NADA demonstrates synergy with the Bcl-2 inhibitor venetoclax, potentially overcoming drug resistance in acute myeloid leukemia (AML).
