Overview

N-hydroxy-nor-L-arginine (nor-NOHA) is a potent, reversible small molecule inhibitor of arginase enzymes, specifically targeting both arginase-1 (Arg-1) and arginase-2 (Arg-2). By inhibiting arginase, it prevents the depletion of L-arginine, thereby increasing its availability for nitric oxide synthase (NOS) to produce nitric oxide (NO). This mechanism is particularly relevant in conditions characterized by endothelial dysfunction, where arginase is often upregulated and competes with NOS for their common substrate. nor-NOHA has been investigated in clinical and preclinical settings for its potential to treat ischemia-reperfusion injury, pulmonary hypertension, coronary artery disease, and obesity-related endothelial dysfunction. It is significantly more potent (approximately 40-fold) than its parent compound, N-hydroxy-L-arginine (NOHA).
