Overview

**n-Methanocarbathymidine** (N-MCT), also known as north-methanocarbathymidine or (north)-methanocarbathymidine, is a synthetic thymidine analog with potent antiviral activity against herpesviruses and orthopoxviruses[6][8]. It acts primarily by inhibiting viral DNA polymerase after being phosphorylated by viral thymidine kinase to its active triphosphate form[8]. N-MCT has demonstrated strong in vitro and preclinical efficacy against herpes simplex virus types 1 and 2 (HSV‑1/HSV‑2), Kaposi’s sarcoma-associated herpesvirus (KSHV), varicella-zoster virus (shingles), and smallpox[8][3][6]. The drug is under development for the treatment of neonatal herpes simplex infection and has received Orphan Drug Designation from the US FDA for this indication[3][5]. Its developer is N&N Pharmaceuticals.
