Overview

N-methyl-pyrrolidone (NMP) is a small molecule traditionally utilized as an industrial solvent and pharmaceutical excipient that is being repurposed as an active therapeutic agent for hematological malignancies. Research, primarily conducted at the Peter MacCallum Cancer Centre, has demonstrated that NMP possesses intrinsic anti-tumor and immunomodulatory activity by acting as a low-potency inhibitor of the bromodomain and extra-terminal (BET) family of proteins, including BRD4. This inhibition leads to the downregulation of key oncogenic drivers such as MYC and IRF4, which are critical for the survival and proliferation of multiple myeloma cells. NMP has been evaluated in Phase I clinical trials for patients with relapsed or refractory multiple myeloma who have exhausted standard treatment options, including lenalidomide and bortezomib.
