Overview

N-oleyl-phosphatidyl-ethanolamine (NOPE) is a naturally occurring phospholipid found in cell membranes and various food sources, currently utilized as a dietary supplement for weight management and the treatment of obesity. Upon oral ingestion, NOPE is hydrolyzed by phospholipase D in the gastrointestinal tract to produce N-oleyl-ethanolamide (NOE) and phosphatidic acid. NOE serves as the primary bioactive component, functioning as a satiety factor that regulates food intake. Its mechanism involves the activation of Peroxisome proliferator-activated receptor alpha (PPAR-alpha) and the indirect modulation of the endocannabinoid system by inhibiting the effects of anandamide on Cannabinoid receptor 1 (CB1). NOPE is frequently commercialized in combination with epigallocatechin-3-gallate (EGCG) from green tea extract, a pairing designed to enhance metabolic rate and improve compliance with calorie-restricted diets. Clinical studies have demonstrated its potential to reduce appetite and improve body composition in overweight and obese populations.
