N-phenyl-1-(phenylsulfonyl)-1H-1,2,4-triazol-3-amine is a small molecule non-nucleoside reverse transcriptase inhibitor (NNRTI) developed by Collaborations Pharmaceuticals for the treatment of HIV-1. This compound belongs to a novel class of 1-sulfonyl-3-amino-1H-1,2,4-triazoles specifically designed to address HIV-associated neurocognitive disorders (HAND) by optimizing brain penetration and minimizing neurotoxicity. The lead candidate in this class, compound 12126065, has demonstrated potent picomolar antiviral activity against wild-type HIV-1 and maintains efficacy against clinically relevant NNRTI-resistant mutant strains. Preclinical studies indicate that the molecule possesses a favorable safety profile with no acute or subacute toxicity and excellent central nervous system penetration.
