Overview

N332-GT5 gp140 is an HIV envelope trimer vaccine candidate designed to induce V3 glycan BG18-class broadly neutralizing antibody (bnAb) precursors. It is a modified Clade A BG505 SOSIP trimer derivative created through computational engineering to maintain the shape of HIV proteins. The vaccine is made from a manmade protein that resembles proteins found on the outside of HIV and is administered with a saponin/MPLA nanoparticle (SMNP) adjuvant to enhance immune response. N332-GT5 gp140 represents a significant improvement over earlier designs, with preclinical studies showing a 14 million-fold improvement in binding affinity to BG18 immunoglobulin precursors compared to initial protein designs. The vaccine cannot cause HIV or AIDS as it is not made from live HIV, killed HIV, or HIV-infected human cells.
