N4 101 is an oral TNF-α inhibitor in pre-clinical development for the treatment of inflammatory bowel disease (IBD), including Crohn’s disease and ulcerative colitis. Developed by N4 Pharma, it utilizes a proprietary non-lipid, non-viral silica nanoparticle delivery system called Nuvec to achieve local delivery of dual-loaded nucleic acids directly to the gut. This targeted approach aims for uptake by macrophages involved in inflammation, with a dual mechanism designed to both reduce excessive TNF-α production and promote the body’s own anti-inflammatory response. The oral formulation seeks to overcome limitations of current IBD therapies—such as injectable antibody-based drugs and small interfering RNAs (siRNAs)—by improving patient compliance and reducing systemic side effects[1][2].
