N501-CYMMAF is a novel near-infrared (NIR) ratiometric fluorescent antibody-drug conjugate (ADC) designed for the theranostic management of metastatic ovarian cancer. It consists of the n501 antibody, which provides high-affinity targeting to ovarian cancer cells, conjugated to the cytotoxic payload monomethyl auristatin F (MMAF) and a cyanine-based ratiometric fluorescent probe via a stimuli-cleavable linker. This "smart" theranostic agent enables targeted delivery of MMAF to inhibit tubulin polymerization while simultaneously allowing for real-time, in situ monitoring of treatment response through ratiometric fluorescence imaging. This dual functionality facilitates the identification of treatment-evading metastatic cell populations and provides insights into tumor recurrence pathways, helping to determine treatment endpoints.
