N57CAR-T is a second-generation, murine-based chimeric antigen receptor (CAR) T-cell therapy designed to target the CD7 antigen. It incorporates 4-1BB and CD3ζ co-stimulatory domains to enhance T-cell activation and persistence. A novel "naturally selected" approach is utilized in its generation to overcome CD7-directed fratricide without requiring gene editing. This therapy is being evaluated for the treatment of relapsed/refractory T-cell acute lymphoblastic leukemia/lymphoma (T-ALL/LBL), particularly in patients with central nervous system (CNS) involvement.
