N6022 is a first-in-class, potent, selective, and fully reversible small molecule inhibitor of S-nitrosoglutathione reductase (GSNOR). GSNOR is an enzyme involved in the metabolism of S-nitrosoglutathione (GSNO) and maintenance of nitric oxide (NO) homeostasis. By inhibiting GSNOR, N6022 restores GSNO levels in the lungs, which may help recover bronchodilatory and anti-inflammatory actions lost in diseases such as asthma and cystic fibrosis. Preclinical studies have shown efficacy in animal models of asthma, chronic obstructive pulmonary disease (COPD), and inflammatory bowel disease. Clinical trials have evaluated its safety and efficacy primarily for acute asthma exacerbations and cystic fibrosis; it has been administered intravenously due to lack of oral bioavailability.
