Overview

N8-acetylspermidine is a naturally occurring acetylated derivative of the polyamine spermidine, produced through both endogenous metabolic pathways and microbial activity in the gut. It has emerged as a significant bioactive metabolite in the context of intestinal homeostasis and inflammatory regulation. Research, particularly involving natural products like lily polysaccharides, has demonstrated that N8-acetylspermidine acts as a key signaling molecule that modulates the cGAS-STING (cyclic GMP-AMP synthase - stimulator of interferon genes) axis. By inhibiting this pathway, N8-acetylspermidine suppresses the production of pro-inflammatory cytokines and alleviates symptoms of colitis. While currently in the preclinical research stage, it represents a potential therapeutic lead or biomarker for inflammatory bowel diseases (IBD), including ulcerative colitis.
