Na-APR-1 (M74) + Alhydrogel is a recombinant protein subunit vaccine candidate developed for the prevention of human hookworm infection. The vaccine consists of a catalytically inactive mutant of Necator americanus aspartic protease 1, known as Na-APR-1 (M74), which is adsorbed onto aluminum hydroxide gel adjuvant (Alhydrogel). The M74 mutation inactivates the enzyme to avoid potential toxicity while retaining immunogenicity. Upon administration, the vaccine induces neutralizing antibodies that target and block the enzymatic activity of native APR‑1 in hookworms, impairing their ability to digest host hemoglobin and feed on blood. This mechanism aims to reduce parasite burden and associated morbidity in infected individuals. Clinical trials have demonstrated that this formulation is safe, well-tolerated, and elicits robust antigen-specific IgG responses in healthy adults[3][5][2].
