Drug intelligence / Profile preview
nab-17-AAG is a **nanoparticle albumin-bound formulation** of 17-allylamino-17-demethoxygeldanamycin (17-AAG), a semi-synthetic derivative of geldanamycin and a potent inhibitor of the molecular chaperone **HSP90 (Heat Shock Protein 90)**. 17-AAG induces the degradation of client proteins involved in cell growth and survival, leading to cell cycle arrest and apoptosis in cancer cells. The **nab- formulation** is designed to improve the solubility and delivery of 17-AAG, potentially enhancing its therapeutic index and allowing intravenous administration. The core mechanism is inhibition of HSP90, leading to destabilization of oncogenic signaling proteins such as AKT, HER2, EGFR, and others. 17-AAG and its formulations have been evaluated primarily for **solid tumors** including hormone-refractory prostate cancer, neuroblastoma, and gallbladder cancer, but clinical studies have not led to regulatory approval.
