This investigational combination therapy consists of the PD-1 inhibitor toripalimab and a triple-drug induction chemotherapy regimen known as TPC, which comprises nab-paclitaxel, cisplatin, and capecitabine. It is currently being evaluated by Sun Yat-sen University in a Phase II clinical trial for the treatment of locally advanced nasopharyngeal carcinoma (NPC). Toripalimab is a humanized IgG4 monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with ligands PD-L1 and PD-L2 to restore T-cell mediated anti-tumor immunity. The chemotherapy components provide synergistic cytotoxic effects: nab-paclitaxel is an albumin-bound nanoparticle formulation of paclitaxel that inhibits microtubule dynamics; cisplatin is a platinum-based agent that induces DNA cross-linking and apoptosis; and capecitabine is an oral prodrug of 5-fluorouracil that inhibits thymidylate synthase, disrupting DNA synthesis. This regimen is typically administered as induction therapy prior to concurrent chemoradiotherapy.
