Overview

Nab-paclitaxel + gemcitabine is an investigational combination chemotherapy regimen currently being evaluated for the treatment of peritoneal metastases from pancreatic cancer. The regimen consists of nab-paclitaxel, a nanoparticle albumin-bound formulation of the taxane paclitaxel that stabilizes microtubules to inhibit mitosis, and gemcitabine, a pyrimidine nucleoside analog that inhibits DNA synthesis and ribonucleoside reductase. In the context of the Nab-PIPAC Phase II trial (NCT05371223), nab-paclitaxel is administered both intravenously and via Pressurized IntraPeritoneal Aerosol Chemotherapy (PIPAC), a laparoscopic delivery method designed to enhance drug penetration into peritoneal nodules, while gemcitabine is administered intravenously. This combination approach aims to address the poor prognosis and limited treatment options for patients with advanced pancreatic cancer involving the peritoneum.
