This is a combination regimen consisting of three agents: - **Nab-paclitaxel** (albumin-bound paclitaxel), a microtubule inhibitor that disrupts cell division by stabilizing microtubules and preventing their depolymerization. It is used as an antineoplastic agent in various cancers. - **S-1**, an oral fluoropyrimidine derivative composed of tegafur (a prodrug of 5-fluorouracil), gimeracil (a dihydropyrimidine dehydrogenase inhibitor), and oteracil (an orotate phosphoribosyltransferase inhibitor). S-1 acts as an antimetabolite to inhibit DNA synthesis in cancer cells. - **Tirelizumab**, a humanized monoclonal antibody targeting programmed cell death protein 1 (**PD-1**), functioning as an immune checkpoint inhibitor to enhance T-cell mediated anti-tumor immunity. This combination is under investigation for the treatment of advanced or metastatic solid tumors, particularly pancreatic ductal adenocarcinoma and other gastrointestinal cancers. The rationale for combining these agents is to leverage cytotoxic chemotherapy with immunotherapy for synergistic anti-tumor effects.
