Nab-paclitaxel + trastuzumab is a combination regimen used primarily in the treatment of HER2-positive breast cancer. Nab-paclitaxel (nanoparticle albumin-bound paclitaxel) is a solvent-free formulation of paclitaxel that avoids hypersensitivity reactions associated with conventional taxanes and does not require premedication. Trastuzumab is a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2/ErbB-2). The combination leverages two distinct mechanisms—nab-paclitaxel disrupts microtubule function to inhibit cell division, while trastuzumab blocks HER2 signaling and mediates immune responses against tumor cells. This regimen has demonstrated efficacy as neoadjuvant therapy for HER2-positive early and locally advanced breast cancer, achieving high rates of pathologic complete response with an acceptable safety profile[1][5][7]. Nab-paclitaxel is developed by Abraxis BioScience (now part of Celgene/Bristol Myers Squibb), while trastuzumab was developed by Genentech/Roche.
